چکیده

PLoS Biology | http://biology.plosjournals.org For half a century, natural products from microorganisms have been the main source of medicines for treating infectious disease. The most important chemical class of these antibiotics, apart from the penicillins, is the polyketides. They are made by the stepwise building of long carbon chains, two atoms at a time, by multifunctional enzymes that determine the chain length, oxidation state, and pattern of branching, cyclisation, and stereochemistry of the molecules in a combinatorial fashion to produce an enormous variety of structures. Recent elucidation of the genetic ‘programming’ of the enzymes has opened a new fi eld of drug discovery based on rationally engineering the enzymes to produce ‘unnatural natural products’ with novel properties. Following the development of penicillin for the treatment of septicemia in the early 1940s, numerous antibiotics were discovered and introduced into medicine. While a fungus makes penicillin, semisynthetic derivatives of which have been a mainstay of antibacterial therapy for decades, most natural antibacterial antibiotics come from a group of soildwelling, fi lamentous bacteria called the actinomycetes, of which Streptomyces is the best-known genus. These organisms make an amazing array of so-called secondary metabolites that have evolved to give their producers a competitive advantage in the complex soil environment, where they are exposed to stresses of all kinds (Challis and Hopwood 2003). The compounds have many functions, but those Primer

Detection and Relation of Polyketide Synthase (PKSs) Genes With Antimicrobial Activity in Terrestrial Cyanobacteria of Lavasan

Background and Aims: Cyanobacteria are considered as favorable source for new pharmaceutical compounds. To date, the majority of bioactive metabolites isolated from cyanobacteria are either polyketides (PKSs) or non-ribosomal peptides. Despite of several worldwide studies on prevalence of PKSs, none of them included the terrestrial cyanobacteria of the Lavasan. Therefore, this study aimed to de...

Database Cracking

This paper talks about the self-oraganized index maintainance approach of database cracking. It provides fundamental knowledge as well as the description of a basic implementation of database cracking. Alongside multiple cracking algorithms, the implementation also contains the code of a cracking index. By combining these two features the implementation allows the execution of range queries on ...

Invited Talk: Encoding Evolvability: The Hierarchical Language of Polyketide Synthase Protein Interactions

Polyketide synthases use an assembly-line mechanism to catalyse the synthesis of antibiotics and other natural products. Each member of a multiprotein complex adds a particular building block to a growing polyketide chain, so the order of the proteins determines the order of the product. In the laboratory, this property has been used to drive combinatorial chemistry; in the bacterial world, pol...

Sensitivity Analysis of Stress and Cracking in Rock Mass Blasting using Numerical Modelling

Drilling and blasting have numerous applications in the civil and mining engineering. Due to the two major components of rock masses, namely the intact rock matrix and the discontinuities, their behavior is a complicated process to be analyzed. The purpose of this work is to investigate the effects of the geomechanical and geometrical parameters of rock and discontinuities on the rock mass blas...

The Origins of Specificity in Polyketide Synthase Protein Interactions

Polyketides, a diverse group of heteropolymers with antibiotic and antitumor properties, are assembled in bacteria by multiprotein chains of modular polyketide synthase (PKS) proteins. Specific protein-protein interactions determine the order of proteins within a multiprotein chain, and thereby the order in which chemically distinct monomers are added to the growing polyketide product. Here we ...
